2,6-二甲基-4-(3-硝基)苯基-1,4-二氢吡啶-3-甲酸乙酯-5-甲酸甲酯不对称合成研究

摘    要:研究以乙酰乙酸甲酯、氨、乙酰乙酸乙酯、3-硝基苯甲醛为原料,在无水乙醇溶剂中通过手性催化不对称合成(R)2,6-二甲基-4-(3-硝基)苯基-1,4-二氢吡啶-3-甲酸乙酯-5-甲酸甲酯.该产物ee值为45%,熔点为163~164℃,产率为85.7%.应用X-衍射、红外光谱、旋光仪、核磁共振光谱对产物的结构进行表征;旋光仪测定该异构体是左旋体;X-衍射晶体结构分析产物是R型.

Abstract:We researched on the asymmetric synthesis of 2,6-dimethyl-4-(3-nitrate yl) phenyl-1,4-dihydro-pyridine-3-ethyl-5-carboxylate by methyl acetoacetate and ethyl acetoacetate and ammonia and 3-nitrobenzaldehyde as the raw material at chiral phase transfer catalyst benzyl bromide in the solvent of anhydrous ethyl alcohol.As a result,the ee value is 45%,melting point is 163~164℃,yield is 85.7%.The structure of the product was characterized via X-diffraction,infrared spectroscopy,polarimetry and nuclear magnetic resonance spectroscopy.Polarimeter measurement revealed that the isomer is levo isomer;and the product is R-type proved by the analysis of X-diffraction crystal structure.
