摘    要:为研究响应面法优化碱性蛋白酶提取野生山核桃籽粕蛋白质的工艺,在单因素试验的基础上考察了加酶量、pH值、提取温度和提取时间等条件对蛋白质提取率的影响。采用Box-Behnken Design试验设计,进行四因素三水平的中心组合试验,建立了蛋白质提取率与各主效因子的二次回归模型方程。结果表明,最佳提取条件为加酶量4.0%、pH12.0、温度47℃、提取时间2.0h。在此条件下,蛋白质提取率可达81.90%。

Abstract:In order to explore the optimum technology of extracting protein with the alcalase protease irom wild walnut cake by response surface methodology,the effect of such factors as alcalase protease content,pH value,extraction temperature and time on the extraction efficiency was studied based on the single factor test. A four factor three level central composite experi- ment was performed with Box-Behnken Design, and quadratic regression model equation between the protein extraction rate and the main effect factor was established. It was showed in the results that the optimum extraction conditions were following as : alcalase protease content 4.0% ,pH 12.0 and extracting 2 hours at 47.0℃. Under these optimum conditions ,the extraction rate of protein is up to 81.90%.
