高表达HER2/neu小鼠乳腺癌肿瘤模型的建立

摘    要:用不同数量的处于对数生长期的D2F2细胞接种于BALB/C小鼠的胸部皮下,建立高表达HER2/neu的小鼠乳腺癌肿瘤模型,并观察小鼠肿瘤的生长特点,应用免疫组织化学染色方法进行鉴定,归纳总结接种细胞数与肿瘤发生率的关系和肿瘤生长对小鼠生活状态的影响。结果显示:在接种细胞数大于5×10^9/L时,BALB/c小鼠的肿瘤模型全部建立成功,免疫组织化学结果显示,肿瘤组织高表达HER2/neu且D2F2细胞接种于BALB/C小鼠的肿瘤模型成功率与接种细胞数成正比。

Abstract:D2F2 cells in logarithmic growth phase were inoculated into BALB/c mice's chest subcutaneous tissue to establish a model of mouse mammary tumors. The growth characteristics of transplanted melanoma in mice were observed and the tumor properties were identified by immunohistochemistry. The relations between the number of D2F2 cells inoculated and tumorigenetic rate, and the influence of tumorigenesis for mice were observed. Results: The tumorigenetic rate in inoculating with over 5 × 10^9/L D2F2 ceils was 100%. The tumorigenetic rate has a positive relation to the number of inoculated D2F2 cells, and is high in BALB/c mice inoculated the D2F2 cells in chest subcutaneous tissue.
