高效液相色谱法测定薄荷中咖啡酸与蒙花苷的含量

摘    要:目的建立薄荷中咖啡酸与蒙花苷的含量测定方法。方法采用HPLC法测定咖啡酸的含量,以Zorbax SB C18为色谱柱;流动相为乙腈-0.1%磷酸溶液(13:87);检测波长为323nm;流速为1ml·min-1;柱温为30℃。采用HPLC法测定蒙花苷的含量,以Zorbax SB C18为色谱柱;流动相为甲醇-0.1%磷酸溶液(48:52);检测波长为334nm;流速为1ml·min-1;柱温为30℃。结果咖啡酸在0.0202~0.4040μg范围内线性关系良好(r=0.9999),平均加样回收率为98.0%(RSD=1.16%)。蒙花苷在0.026~0.520μg范围内线性关系良好(r=0.9999),平均加样回收率为99.0%(RSD=1.22%)。结论方法简便、重复性好,为薄荷的质量控制研究提供了一定的参考。

Abstract:Objective To set up the determination method of caffeic acid and linarin content in Herba Menthae. Methods Using HPLC to determine the cafffeic acid.Set chromatography column as Zorbax SB C18 mobile phase as acetonitrile-0.1% phosphoric acid(13:87);detection wavelength was 323nm;flow rate of 1ml·min-1; column temperature was 30℃.Using HPLC to determine the linarin content,set chromatography column as Zorbax SB C18;mobile phase as methanol-0.1% phosphoric acid (48:52);detection wavelength was 334nm;flow rate of 1ml﹒min-1;column temperature was 30℃.Results In the range of 0.0202~0.4040μg,caffeic acid showed good linear relationship(r=0.9999),the average recovery was 98.0%(RSD=1.16%).Linarin in the range0.026~0.520μg also showed good linear relationship(r=0.9999),the average recovery was 99.0%(RSD=1.22%).Conclusion HPLC is a simple,reproducible method which provides valuable reference for the study of Herba Menthae quality control.
