多酶分步水解法提高豌豆纤维纯度的研究

摘    要:以豌豆粉丝废水分离渣为原料,对豌豆纤维的提取过程进行研究.通过耐高温α-淀粉酶、糖化酶、蛋白酶多酶分步水解的方法,去除豌豆渣中的淀粉和蛋白质.结果表明,耐高温α-淀粉酶添加量1.2%,反应温度为90℃,反应时间为60 min的条件下,豌豆膳食纤维的提取率最高;糖化酶加酶量为4.5%,反应温度为60℃,反应时间为110 min,pH为5.0的条件下,可达到良好的水解效果;蛋白酶的加酶量为1.0%,反应温度为55℃,反应时间为80 min,pH为8.5的条件下,酶解效果最佳.此时豌豆粉丝废水分离渣的膳食纤维含量可达到94% 以上.

Study on Improving the Purity of Pea Fiber by Multi Enzyme Hydrolysis Steps

Abstract:The extraction process of pea fiber was studied from pea vermicelli production wastewater separation residue as raw material.The starch and protein contained in the pea residue are removed by multi enzyme hydrolysis steps with heat-resisting a-amylase,saccharifying enzyme and protease.The results show that when the addition of heat-resisting a-amylase is 1.2%,enzymatic hydrolysis at 90℃ for 60 min,pH is 7.0,Pea dietary fiber has the highest extraction rate.Glucoamylase can achieve a good hydrolysis effect under the conditions that the amount of enzyme added is 4.5%,the reaction temperature is 60℃,the reaction time is 110 min,and the pH is 5.0.When the addition of alkaline protease is 1.0%,enzymatic hydrolysis at 55℃ for 80 min,pH is 8.5,the enzymatic hydrolysis is best.Under these conditions,the pea fiber content can reach more than 94 %.
