碱提法提取大球盖菇多糖工艺及其清除氧自由基研究

摘    要:为获得大球盖菇碱性多糖的最佳提取工艺,本文采用水浴碱提法提取多糖,通过单因素和正交实验对多糖提取的影响因素(提取温度、碱液浓度、时间)进行分析,优化多糖提取工艺并对提取的多糖进行抗氧化活性测定。结果表明,提取温度显著影响多糖得率,最佳工艺条件为提取温度85℃、碱液浓度0.03 mol·L-1、时间2 h。大球盖菇碱性多糖具有较强的抗氧化能力,对在最佳工艺条件下得到的大球盖菇碱性多糖进行活性氧自由基清除研究,发现多糖浓度在0.8 mg·mL-1时,对氧自由基的清除率可达到80.12%。

Abstract:In order to obtain the best extraction process of Stropharia rugoso-annulata alkaline polysaccharides,this article uses water bath alkaline extraction to extract polysaccharides,and analyzes the influencing factors(extraction temperature,lye concentration,time)of polysaccharide extraction through single factor and orthogonal experiments.Optimize the extraction process of polysaccharides and determine the antioxidant activity of the extracted polysaccharides.The results show that the extraction temperature significantly affects the yield of polysaccharides.The best process conditions are the extraction temperature of 85℃,the concentration of lye 0.03 mol·L-1,and the time of 2 h.Stropharia rugoso-annulata alkaline polysaccharide has strong antioxidant capacity.The Stropharia rugoso-annulata alkaline polysaccharide obtained under the optimal process conditions was studied for active oxygen free radical scavenging,and it was found that the polysaccharide concentration was 0.8 mg·mL-1,The scavenging rate of oxygen free radicals can reach 80.12%.
