橡胶草叶片愈伤组织诱导研究

摘    要:以橡胶草的幼嫩叶片为试验材料,对外植体愈伤组织诱导的优化条件进行了试验研究。试验以MS为基本培养基,以0.1%氯化汞溶液为消毒液,以6-BA和2,4-D为诱导激素,以愈伤组织诱导率和生长状况为筛选指标,不同消毒时间和不同浓度的6-BA和2,4-D对橡胶草愈伤组织诱导的影响进行了试验研究。试验结果表明:6-BA和2,4-D对诱导橡胶草叶片愈伤组织均有影响,最优培养基为MS+6BA 2.0 mg/L+2,4-D 1.0 mg/L,诱导率达83.3%,且愈伤组织生长状况良好。

Study on Leaf Callus Induction of Taraxacum kok-saghyz

Abstract:Taking young leaf of Taraxacum kok-saghyz as experimental material, optimum inducing conditions of callus of explant is studied. Taking MS as the basic base, 0.1% content of HgCI solution as disinfectant, 6-BA and 2, 4-D as inducing hormones, inducing rate and growth state as screening indexes, impact of different sterilizing time and different content of 6-BA and 2, 4-D on inducing callus is studied. Results indicate that 6-BA and 2,4-d all have impacts on leaf callus of Taraxacum kok-saghyz, the best culture base is MS + 6BA 2.0 mg/L + 2,4- 1.0 rag/L, with inducing rate of 83.3%, and growth state of callus is well.
