摘    要:目的]改善明亮发光杆菌细胞通透性、提高Microtox对有机污染物生物毒性检测的灵敏度。方法]以乙二胺四乙酸二钠(EDTA-Na2)处理明亮发光杆菌,用疏水荧光探针N-苯基-1-萘胺(NPN)检测明亮发光杆菌通透性的改变,应用发光细菌急性毒性检测技术(Microtox)检测有机化合物在通透性改变前后的50%发光抑制浓度(EC50)。结果]加入EDTA后菌体对NPN的吸收系数明显增加,40mM的ED-TA可使明亮发光杆菌对NPN的吸收系数增加47.7%。未经EDTA处理时,1,10-二氮杂菲、1,4-对位苯醌、双酚A、4-异丙基苯酚经Microtox检测的EC50值分别为181.2、0.071、37.6、0.79mg/L,处理后分别降为115.1、0.054、26.24、0.603mg/L。结论]EDTA可以提高明亮发光杆菌对有机化合物的通透性,使Microtox检测生物毒性的灵敏度提高。

Abstract:Objective]To improve the permeability of Photobacterium phosphoreum and the sensitivity of Microtox to toxic organic compounds.
