体外培养兔成骨细胞的改进方法

摘    要:为了提高体外培养兔成骨细胞的效果,实验选用了出生15天幼兔的颅盖骨,将酶消化法和组织块法相结合,体外分离培养了成骨细胞并对其进行了鉴定.倒置显微镜及Giemsa染色观察培养细胞的形态及生长特征,碱性磷酸酶(ALP)和茜素红染色分别显示培养细胞的碱性磷酸酶活性和矿化能力,结果显示培养细胞具有体内成骨细胞的形态学特征和生物学行为.改进的兔颅盖骨成骨细胞体外培养方法是成功的,且具有操作简单、费用低以及细胞几乎不受损伤的优点.

Abstract:To improve the effect of rabbit osteoblast culture in vitro,the osteoblasts isolated from the calvarial bone of newborn rabbit for 15 days are cultured with the combined method of enzyme digestion and tissue mass culture.The acquired cells are assayed for osteoblastic properties by cell morphology,alkaline phosphorase(ALP) activity and production of mineralized matrix.The result shows that the cells have the morphologic characteristics and some biological behaviors of osteoblast.So,the improved culture method of osteoblasts from newborn rabbit cranium,which has the advantages of simple operation, low cost and no impairment to the cell,proves to be very successful.
