表面活性剂处理炭疽杆菌的原子力显微镜研究

摘    要:利用AFM观察经表面活性剂处理的炭疽杆菌繁殖体形态,研究不同表面活性剂对细菌的作用。用同一浓度的非离子型表面活性剂NP-40、Span-20、Span-80、Igepal CA-210、Igepal CA-520、Igepal CA-630,对完整的炭疽杆菌菌株进行处理,经AFM成像,观察不同表面活性剂对菌体形态的破坏程度。结果不同的表面活性剂对菌体破坏程度不一,但菌体形态均被破坏,表面形态变得不规则,菌体内部物质不均匀分布。表面活性剂作用后的炭疽杆菌微观形貌与正常菌体相比,因表面活性剂亲水亲油平衡值(HLB)不同而不同,随着HLB值的增加,菌体的破坏程度和表面粗糙度也在增加。本实验为研究炭疽杆菌对不同试剂的抗性提供了形态学方面的基础,也为不同表面活性剂作为消毒剂方面的研究提供了形态学基础,可提供一种较为温和的细菌裂解条件。

Abstract:In the article, the interaction of surfactant and morphous of Bacillus anthracis (B. anthracis) was discussed. After B. anthracis propagule were treated with NP - 40, Span - 20, Span - 80, Igepal CA - 210, Igepal CA - 520 and Igepal CA - 63, the destruction of bacterial structure was observed. The morphous of B. anthracis was differently destroyed by different surfactants. All the processed bacteria cells became irregular in shape. Consequences display the height image, phase image and three -dimension image, and especially acquire the microdomains image of the B. anthracis which has been changed by nonionic surfactant before and after. The morphological characters of B. anthracis have been acquired through its AFM images. The morphous of B. anthracis bacteria cells treated with surfactants has greatly changed and quantifiable indexes are different because of dissimilar surfactants HLB.
