摘 要: | 采用超高效液相色谱-四极杆/静电场轨道阱质谱建立了具有提高缺氧耐受力功能的保健食品中那非类物质的测定方法。样品经甲醇提取,采用Waters Acquity UPLC BEN T3色谱柱(2.1 mm×100 mm,1.8μm)分离,0.1%甲酸水溶液和乙腈溶液为流动相进行梯度洗脱。采用Full MS/dd-MS2扫描模式,结合精确分子质量数、保留时间和二级特征碎片离子对目标化合物进行筛查与确证。结果表明,90种目标化合物在0.1~10 mg/kg范围内线性关系良好(r^2>0.99);定量限(LOQ)为0.01~5 mg/kg,回收率为61.8%~119.1%,相对标准偏差(RSD)为1.2%~12.9%。该方法简单、准确、快速,适用于具有提高缺氧耐受力功能的保健品中90种那非类物质的定性筛查和定量分析。

Abstract: | An ultra-performance liquid chromatography–quadrupole/exactive orbitrap mass spectrometry was developed for determination of 90 nafils in health care products to improve hypoxia tolerance in plateau area.The samples were extracted by methanol,and separated on a Waters Acquity UPLC BEN T3(2.1 mm×100 mm,1.8μm)column using a binary solvent system composed of 0.1%formic acid aqueous solution and acetonitrile solution by gradient elution.The analytes were precisely screened and confirmed with the precision mass,retention time and two-stage ion mass spectra in Full MS/dd-MS^2 scan mode.The result showed that good linear relationships for 90 analytes in range of 0.1-10 mg/kg(r^2>0.99).The limits of quantitation(LOQ)were in range of 0.01-5 mg/kg,The recoveries ranged from 61.8%to 119.1%with relative standard deviations(RSD)of 1.2%-12.9%.The method was simple,sensitive and accurate,and was suitable for the rapid screening and confirmation of 90 nafils in health care products to improve hypoxia tolerance in plateau area. |
