基于GC-MS技术对钩吻碱食物中毒的应急检测

摘    要:目的:对一起食物中毒事件中毒因子进行应急检测,为患者救治及做好此类中毒的检测提供参考和依据。方法:样品经10%NaOH溶液超声提取10 min,三氯甲烷液液萃取,微孔滤膜净化,采用气相色谱-串联质谱Scan模式全扫描,Nist17谱库检索定性,比对标准品和样品质谱图离子丰度比确证、SIM模式下外标法定量。结果:洗胃液、呕吐物、钩吻根猪脚汤3份样品中检出钩吻碱,浓度分别为:0.075 mg·kg-1、0.384 mg·kg-1、6.780 mg·kg-1。结论:该起食物中毒因子是钩吻根猪脚汤中的钩吻碱,气相色谱-串联质谱仪在食物中毒应急检测中可以快速准确对中毒因子定性定量,能应对食物中毒突发性和中毒因子的不确定性。

Emergency Detection of a Food Poisoning by Gelsemine by Gas Chromatography Tandem Mass Spectrometry

Abstract:Objective:To conduct emergency detection of poisoning factors in a food poisoning incident,to provide a basis for the treatment of patients,and to provide a reference for the detection of such poisoning.Methods:Samples were extracted with 10%NaOH solution ultrasonically for 10 minutes,chloroform was extracted with liquid-liquid,and cleaned by microporous membrane.Gas chromatography-tandem mass spectrometry Scan mode was used for full scan,Nist 17 library search and qualitative,and the standard and sample mass spectra were compared.Fig.Confirmation of ion abundance ratio and quantification by external standard method in SIM mode.Results:Gelsineine was detected in three samples of gastric lavage fluid,vomit,and Gelsemium root and pig’s feet soup.The concentrations were 0.075 mg·kg-1,0.384 mg·kg-1,and 6.780 mg·kg-1,respectively.Conclusion:The food poisoning factor is the gellenine in Gelsemium root pig’s feet soup.Gas chromatography-tandem mass spectrometry can quickly and accurately qualitatively and quantify the poisoning factor in food poisoning emergency detection,and it can deal with sudden food poisoning and poisoning.The uncertainty of the factor.
