高效液相色谱串联质谱分析黄芩水煎液的有效成分

摘    要:目的采用高效液相色谱串联质谱(HPLC-MS/MS)联用技术,对黄芩水煎液中的有效成分进行初步分析。方法采用AgilentZORBAXSB-C18色谱柱,以乙腈和水(含体积比0.5%甲酸)为流动相,采用梯度洗脱,流速为0.4mL/min,ESI-MS采用负离子模式。通过HPLC-MS/MS在线测定化合物的一级和二级质谱信息,结合文献报道,对黄芩水煎液的有效成分进行分析鉴别。结果结合文献报道和质谱数据,21个成分通过串联质谱进行了结构鉴定,4个通过对照品得到了确证。结论高效液相色谱串联质谱法是鉴定黄芩药材化学成分的有效方法。

Abstract:Objective To identify the active components in Radix Scutellariae by high performance liquid chromatography coupled with tandem mass spectrometry(HPLC-MS/MS).Methods The separation was performed on an Agilent ZORBAX SB-C18 column with gradient elution of acetonitrile and water(containing 0.5% formic acid)as mobile phase,the flow rate was 0.4 mL/min.On-line ESI-MS/MS in negative mode was used to obtain the MS1 and MS2 data of the analytes.Results According to the literature and high-resolution data,21 compounds were identified by HPLC-MS/MS and 4 of them were confirmed by a comparison of their tR,MS data with those of the reference substances.Conclusion HPLC-MS/MS is demonstrated as a powerful tool for the rapid identification of the chemical constituents in Radix Scutellariae.
