HPLC-ELSD法测定3种广式凉果中的返砂糖

摘    要:本文建立了高效液相色谱-蒸发光散射法检测广式凉果返砂糖中单糖和双糖的方法。采用Shodex Asahipak NH2P-504E(4.6 mm×250 mm,5μm)色谱柱,流动相为乙腈-水(70∶30,V/V),流速为1 mL·min-1,柱温为25℃,蒸发光检测器以N2为载气,压力为350 kPa,漂移管温度为60℃。结果表明,蔗糖、葡萄糖和果糖三者达到基线分离,3种糖在1~10 mg·mL-1的浓度范围内线性关系良好,加标回收率在95.12%~104.65%。3种凉果返砂糖中未检测到蔗糖,主要含有葡萄糖和果糖。

Determination of Granulated Sugar in Three Kinds of Guangdong-Style Preserved Fruits by HPLC-ELSD

Abstract:This paper established a high performance liquid chromatography-evaporative light scattering method for the detection of monosaccharides and disaccharides in granulated sugar of Guangdong-style preserved fruits.Using Shodex Asahipak NH2P-504E(4.6 mm×250 mm,5μm)chromatographic column,the mobile phase is acetonitrile-water(70∶30,V/V),the flow rate is 1 mL·min-1,and the column temperature is 25℃,The evaporative light detector uses N2 as the carrier gas,the pressure is 350 kPa,and the drift tube temperature is 60℃.The results showed that sucrose,glucose and fructose reached the baseline separation,the three sugars had a good linear relationship within the concentration range of 1~10 mg·mL-1,and the spiked recovery rate was 95.12%~104.65%.Sucrose is not detected in granulated sugar of three kinds of Guangdong-style preserved fruits,and it mainly contains glucose and fructose.
