尿素包合法纯化α-亚麻酸方法的研究

摘    要:以亚麻籽油为原料对尿素包合法纯化α-亚麻酸的工艺条件进行研究.采用单因素实验考察包合时间、包合温度、尿素与脂肪酸质量比、甲醇体积与尿素质量比对提取α-亚麻酸的影响,同时采用响应曲面法分析建立二次回归模型,对影响因素包合时间、尿素与脂肪酸质量比、甲醇体积与尿素质量比进行优化组合.以产物中α-亚麻酸质量浓度为指标,确定了尿素包合法提取α-亚麻酸的最佳工艺条件,即包合时间12.80 h,尿素与脂肪酸质量比为2.82,甲醇体积与尿素的质量比为2.44 mL/g,该工艺制备α-亚麻酸质量浓度为179.96μg/mL.采用紫外光谱检测将样品与标准品进行对照,确定样品为α-亚麻酸.

Abstract:α-linolenic acid purification process conditions were studied by urea inclusion in flaxseeds oil as the raw materials .Single factor experiment were carried out to determine the best conditions for time of inclusion , temperature of inclusion , the mass ratio of urea and fat-ty acid, the ratio of methanol volume and urea quality on the extraction of α-linolenic acid and response surface methodology establish the quadratic regression model analysis .The time of inclusion , the mass ratio of urea and fatty acid , the ratio of methanol volume and urea quality which were optimized were combined .The optimum conditions of extracting α-lino-lenic acid were determined by the target of α-linolenic acid and the product yield index . The time of inclusion , the mass ratio of fatty acid and urea , methanol volume and the quality of urea ratio was 12.80, 2.82, 2.44 respectively.Preparation of this process, the mass con-centration of α-linolenic acid was 179.96 μg/mL, The sample were compared with the standard by UV spectrophoto -meter and determine the sample of α-linolenic acid .
