口蹄疫病毒3AB克隆表达及其抗体间接ELISA检测方法建立与应用研究

采用重叠PCR方法人工合成3AB基因片段,将其克隆到PET-28a＋载体,转化导入宿主菌BL21（DE3）中成功表达,获得大小约为33Ku的重组表达蛋白。以纯化的重组表达蛋白为抗原,建立了检测猪FMDV3ABNSP抗体的间接ELISA方法。将本方法与进口口蹄疫非结构蛋白3ABC阻断ELISA试剂盒进行比较,结果显示,本方法可以用于区分猪口蹄疫自然感染动物与疫苗免疫动物。     

鹅细小病毒延边株VP3基因原核表达质粒构建

本试验根据GenBank已公布的鹅细小病毒B株基因序列,设计了一对扩增VP3基因的特异性引物,对鹅细小病毒延边分离株基因组DNA进行了PCR扩增,得到了约为1500bp的VP3基因片段,将其回收纯化后与pMD-18T simple载体连接,然后进行克隆,经双酶切分析、PCR鉴定及测序,验证了所克隆的基因是鹅细小病毒VP3基因.用EcoR Ⅰ和Xho Ⅰ双酶切该片段并回收该片段,将此基因片段克隆至相同酶切回收后的PGEM-4T-1原核表达载体中,获得重组质粒PGEM-VP3,经PCR鉴定,限制性内切酶双酶切分析,证实了重组质拉的正确性.     

五种血清型柯萨奇病毒的实时荧光RT-PCR检测方法研究

在柯萨奇病毒基因组VP1区设计引物和探针,分别建立了特异性检测5种血清型（A9、A16、B2、B3和B5型）柯萨奇病毒（Coxsackievirus）基于MGB探针的实时荧光RT-PCR方法。通过构建的分别适于5种血清型病毒检测的质粒标准分子对建立的检测体系进行了灵敏度分析,确定5种血清型柯萨奇病毒检测体系的检测下限均为2拷贝质粒标准分子DNA。     

应用DNA条形码技术快速鉴定入境船舶上截获的昆虫蛹

[目的]利用DNA条形码技术快速准确鉴定从入境船舶上截获的未知双翅目昆虫蛹。[方法]将截获的5只蛹,1只蛹直接冷冻,4只孵化出成蝇;对蛹、蛹壳及孵化出的成蝇进行拍照留档。分别从蛹、蛹壳和孵化成蝇中提取基因组DNA;采用动物DNA条形码通用引物（LCO1490和HCO2198）扩增目的片段,并进行纯化、克隆测序和序列分析,与NCBI及BOLD中的序列进行比对,构建NJ树;对孵化成蝇进行形态鉴定。[结果]所检测的蛹、蛹壳以及孵化成蝇的DNA条形码序列与NCBI和BOLD中大头金蝇（Chrysomyia megacephala）的序列相似度达100%;孵化成蝇与大头金蝇的形态特征相符。[结论]根据测定结果,判定截获的未知双翅目昆虫蛹为大头金蝇的蛹;证明DNA条形码技术是一种鉴定未知昆虫蛹的有效手段。对于口岸截获的外来未知昆虫蛹、成虫残肢甚至蛹壳均可直接采用DNA条形码技术进行种类鉴定,大大缩短鉴定周期。     

基因表达调控与人类疾病

DNA甲基化、组蛋白修饰及RNA分子的作用可在不同层面影响DNA分子的表达,其中任何环节出现错误都会导致不同的表达错误,从而引发人类疾病。如果我们能控制DNA的表达,将可以使癌症、病毒引发的疾病(如肝炎、艾滋病)、血液疾病等得到治愈。这篇文章中我们将涉及到小分子RNA,核糖开关,假基因,DNA组蛋白的修饰及DNA的甲基化对基因表达的调控。     

泛素结合酶UFC1基因的原核表达载体构建

克隆人的泛素结合酶UFC1(ubiquitin-fold modifier conjugating enzyme 1)基因,并为实现UFC1基因的原核表达、纯化及多克隆抗体制备做准备。方法:采用RT-PCR方法从宫颈癌Hela细胞中扩增UFC1基因全长cDNA,双酶切后克隆到原核表达载体pGEX-4T-2中,构建并鉴定pGEX-4T-2-UFC1质粒。结果:双酶切鉴定和测序分析均证实,已成功构建pGEX-4T-2-UFC1重组载体。结论:成功构建泛素结合酶UFC1基因的原核表达载体,为在原核系统中表达UFC1重组蛋白,并制备多克隆抗体奠定了基础,也为进一步研究UFC1基因功能提供了工具。     

Lea14基因原核表达载体的构建

Lea基因编码的LEA蛋白作为一种晚期胚胎丰富脱水保护蛋白，当植物处于逆境状态下时，其能够高水平的表达，通过降低其脱水的程度来达到抵御逆境的功能。为了进一步研究Lea14基因在逆境反应中的功能，本研究以拟南芥cDNA为模板，扩增出Lea14基因，再将Lea14基因克隆到原核表达载体pET-28a中，获得了Lea14基因的原核表达载体pET-28a-Lea14，测序结果表明：pET-28a-Lea14载体构建成功，可用于后续蛋白表达研究工作。     

免疫琼脂扩散法检测肉制品中的掺假蛋白

制备肌肉冻融水浸液为兔体免疫抗原,28天后获得特异性的高滴度抗血清。运用免疫琼脂扩散法检测掺假蛋白,以标准抗原与抗血清的沉淀线为比较基准,将样品与抗血清所生的沉淀线与之比较,根据琼脂板上二种沉淀线距中心孔(抗血清孔)的距离,判断样品掺假的程度。
